On Windows 10 systems, the file could be in any one of the 4 locations (you can copy-paste each path in the RUN window or the address bar in Windows Explorer):
%ProgramData%MicrosoftWindowsStart MenuPrograms
%ProgramData%MicrosoftWindowsStart MenuProgramsStartUp
%AppData%MicrosoftWindowsStart MenuProgramsStartup
%AppData%MicrosoftWindowsStart MenuPrograms
One final but unlike location that might be worth checking: C:UsersAll UsersStart MenuProgramsStartUp
If the Desktop.ini file is still opening on your screen, first close it, then delete any Desktop.ini at any of these locations to Recycle Bin (initially)
Then restart your computer to check. If after 15-20 seconds Desktop.ini didn't pop-up again, all should be well!
Feel free to empty out the Recycle Bin and change: View > Options > View > check the box next to "Hide protected operating system files (Recommended)", and you are done.
Though not certain, I believe installation or running a dated 32-bit Windows app is likely to have caused this situation. If it continues to occur, focus on what you installed or changed right before the issue surfaced.

Go to the Notepad window that has the desktop.ini file.
In the menu, click File -> Save As.
In the “Save as type”, select “All Files”.
You will see the desktop.ini file in the list, right click, and select Delete.
Cancel the Save As, and close the Notepad window.
Now it will not appear anymore when you reboot.
This is stupid
i found almost 35 desktop.ini file all i had to do was search it in the search box for the disk, should i delete all of them?

didnt work. in control panel I already had that ticked on and start up folder is empty… notepad still comes on at the start on win10 🙁
